titleOfInvention PROCEDURE FOR THE PREPARATION OF CARBAMATE DERIVATIVES OF PHYSOSTIGMINE FROM ESERETOLS AND PROCEDURE FOR OBTAINING AN INTERMEDIARY SUCH PROCEDURE
abstract The product that is prepared has the formula (I) where R is alkyl; R1 is hydrogen, alkyl, cycloalkyl, cycloalkyl alkyl, bicycloalkyl, aryl or arylalkyl; R2 is alkyl, cycloalkyl, cycloalkyl alkyl, bicycloalkyl, aryl, or arylalkyl; or R1 and R2, taken together with the nitrogen atom to which they are attached, form a group of formula (Ia) where Y is hydrogen or alkyl and Z is hydrogen, alkyl, halogen, alkoxy, halogen or trifluoromethyl, and month 0, 1 , or 2, or a pharmaceutically acceptable salt thereof. The procedure consists of (a) contacting a compound of formula (II) as defined herein with reinforced hydrogen bromide to obtain a compound of formula (III) as defined herein; Contacting the reaction mixture containing a compound of formula (III) with: (1) an isocyanate of formula R1NCO or (2) a compound of formula (IV) as defined to obtain a compound of formula (V) as defined herein and contacting the reaction mixture containing the compound of formula (V) with an amine of formula R1R2NH here in the presence of a carboxylic acid of formula R5COOH and forming and isolating the product of formula (I). The preparation of the intermediary (III) is also described.
